I believe multipass scanning reduces noise in shadow regions, but doesn't actually bring out more detail.
For best shadow detail, scan using 16/48 bit precision and play with the exposure adjustment.
I use the color curves transformation in Picture Window Pro to boost shadow contrast-- it can help bring out a lot of detail. Photoshop has an equivalent transformation.
